AB OBJECTIVE: Hitherto the morphological studies on the HIV had revealed
only the high-electron dense materials just under the plasma membranes
at budding sites. To distinguish hidden features of the viral proteins,
combined-technique of immuno electron microscope (IEM) and detergent
treatment was applied to the HIV-infected cells. METHODS: Two cell lines
(TALL-1/KB-1 and Molt-4/HIV-1) were used. The samples for pre-embedded
IEM were treated with 0.02% saponin. RESULTS: The viral proteins (gp120,
p24, and p17) were visualized on the cytoskeletal net works. These
proteins were synthesized by the attached ribosomes on rERs and
cytoskeletons, and then transferred via cytoskeletons to the plasma
membrane, where env protein (gp120) lifted up the membrane and then
sticked into it to construct the viral envelope. gag proteins (p24/p17)
were composed according to the arrangement of env protein. DISCUSSION
AND CONCLUSIONS: Combined-technique of IEM and stereo observation was
useful to reveal the distributions of unobserved viral proteins.
Different from usual protein synthesis, the viral proteins of the HIV
were synthesized on the attached ribosomes on rERs and cytoskeletons
with no relations to ERs or Golgi apparatus. These results would also
suggest several informations for other Retroviruses.
